From API to Nano Drug Product / Part 2: Preparation and Characterization of Nanosuspensions, Loaded Granules, and Pellets*Part 1 see Pharm. Ind. 2026;88(6):470–474. · Shi S, Schmidt A, Eugster D, Grave A · Glatt Pharmaceutical Services und Willy A. Bachofen und Glatt Pharmaceutical Services
Particle Size Morphology Redispersibility Dissolution Stability To establish a stable nanomilling process, 2.5 % Kollidon® VA 64 was selected as a stabilizer together with 0.25 % SDS in the naproxen (nano)suspensions [ 13 , 16 ]. This combination played an important role in the successful nanomilling of naproxen nanoparticles as well as in maintaining nanosuspension stability during storage prior to fluid bed processing (approx. 3 d). For the determination of basic process parameters, initial experiments were conducted on a small-scale wet bead mill with a batch size of 2 kg. The parameters were then scaled up to a manufacturing-scale wet bead mill with an 8 kg batch ...

Wet Bead Milling Fluid Bed Top-Spray Granulation Drug Layering Pellets Active pharmaceutical ingredients (APIs) with poor aqueous solubility, especially when associated with limited dissolution rates, pose a significant challenge in pharmaceutical development, as they limit bioavailability and therapeutic efficacy [ 1 ]. This issue has become more critical, affecting up to 70 % to 90 % of certain newly identified APIs [ 2 ]. These APIs, due to their low solubility, are typically categorized as Biopharmaceutical Classification System (BCS) Class II or Class IV [ 3 ]. The solubility issue is particularly pertinent for oral administration, which remains the most prevalent and desired route for drug delivery. ...
